Inspirations

Scientific part

Our project is based on the construction of a bacteria lemon that will look like lemon but made by only Escherichia coli modified to produce lemon scent and change colour from green to yellow. Thus, we will achieved our project by genetically modifying Escherichia coli to be free of any unpleasant odour mutating genes involved, by introducing genes for the production of lemon scent and amplifying this pathway and finally adding a yellow-bleu chromoprotein which its expression is controlled by tRNA suppressor for the colour changing.

Remove the bad smell of E.coli

Our lemon should smell like a lemon. Using Escherichia coli as a chassis, that everybody knows has a foul odor, we have to remove genes involved in this particular phenotype. tnaA, which is one of that gene, is required for the degradation of tryptophane into indole the main element that provide that smell. We use a phage transduction method to reach our goal.

Lemon Scent

The particular odor of lemon comes from 3 main monoterpenes: limonene, beta-pinene and geraniol. Cloning these 3 monoterpene-synthase into our bacteria, we allow it to produce this fragrance. It requires also to improve the production of the precursor GPP using the mevalonate-pathway from Eukaryota organism.

Lemon appearance and Ripening

Since vision is one of the most important senses in humans, we want the lemon look like as real as possible. To achieve this, we planned to make our lemon look green or yellow, as a real one. Furthermore, we also wanted to simulate the ripening process of the lemon by changing its color gradually from green to yellow.
