Scaffold GSP
The scaffold GSP was designed in silico with GS linker groups between the single domains. Additionally, a BamHI restriction site was added at the C-terminal of the protein. Its sequence was codon optimized to improve the ability of the scaffold to be expressed in E. coli and then synthesized via solid phase DNA-synthesis. For the generation of a new Biobrick the sequence was cloned into pSB1C3.
