Colony PCR was done to check the right clones.
 
Positive ligated products were inoculated in LB media containing respective antibiotics.
 
K1033222 was ligated to K081009-B0015 and B0034 was ligated to E0040-B0015 construct.
 
Transformed into E.coli.
 
Colony PCR to check the size.
