Export Systems
Preparation of fsr system for LC/MS analysis:
- started vacufuging pAA001 acetonitrile samples (negative controls).
- Colony PCR was run on 8 more colonies picked from pTG005-noFLAG plate (RTH reaction run last week).
- Ran the PCR products on a gel.
- Set up 6 LB-CARB liquid cultures corresponding to the promising colonies, as determined by the colPCR

- Performed another QuickLigation reaction using yesterday's round-the-horn PCR product.
- Ligation reaction product was transformed into JM109.
- Picked six colonies from yesterday's transformations (RTH) and restreaked on a new plate & started overnight liquid cultures.

- The 6 liquid cultures prepared yesterday were miniprepped and shipped for sequencing.
