June 10,2014 we mini prepped i5.9.1 which was RBS and also i5.9.2 which was promoter. We then did a digestion and gel electrophoresis and Purification for the promoter using a 2 log ladder as seen in figure:6.10.1. Finally we Ligated and Transformed P-Rbs-eCFP,P-Rbs-YG,P-Rbs-eYFP,P-Rbs-redch., RBS-bluechromo., and a negative control.
