Export Systems
Preparation of fsr system for LC/MS analysis:
  • started vacufuging pAA001 acetonitrile samples (negative controls).
  • Colony PCR was run on 8 more colonies picked from pTG005-noFLAG plate (RTH reaction run last week).
  • Ran the PCR products on a gel.
  • Set up 6 LB-CARB liquid cultures corresponding to the promising colonies, as determined by the colPCR

  • Performed another QuickLigation reaction using yesterday's round-the-horn PCR product.
  • Ligation reaction product was transformed into JM109.
  • Picked six colonies from yesterday's transformations (RTH) and restreaked on a new plate & started overnight liquid cultures.

Tuesday, 8/19/14

Export Systems
  • The 6 liquid cultures prepared yesterday were miniprepped and shipped for sequencing.

  • Performed colony PCR on the round-the-horn colonies obtained from Sunday's & Monday's transformations.
  • Sent promising colPCR products for sequencing.

Preparation of our powerpoint presentations for SURF seminar day (see Thursday's entry).

Thursday, 8/21/14

Today is Summer Seminar Day (for SURF, our official summer research program)! All of us gave ~20-minute presentations on our individual subprojects.
