Some plates didn’t have individual colonies; just smears. This is likely due to the fact that we added too little antibiotic to our plates. Instead of adding 250 µl of CM, we added 25 µl. So, we threw out our plates and made more.

We also designed Gibson primers to put the BioBricks in the appropriate plasmids.

We attempted to replate the BioBricks that we couldn’t pick. This time, we made serial dilutions so that the final concentration relative to the number of cells we got from the stab was 1:10,000.
