Today’s result was clear: Having tried the transformation for three times we found out that E. coli RR1 suffered a mutation that made it resistant to chloranphenicol, selectable marker from the pSB1C3. We have lost time and lots of microliters from the diluted DNA from the 2013 Parts kit! We lost a precious amount of time but the result was gratifying.

After having this sorted out we have discussed the best methodology to make DH5a chemical competent. We decided to use the methodology given by Professor Spartaco.

We have made a pre-inoculum in liquid LB media !
