    PCR step 3 : Vtot = 50µL

    10µL phusion buffer 5X
    1µL dNTP 10mM
    0,5µL phusion polymérase
    2,5µL DMSO
    2,5µL Primer 1 à 10mM
    2,5µL Primer 8 à 10mM
    10µL 3° ou 3°°
    21µL H2O

    PCR Parameter:
    98°C 5 min
    35 cycles : 98°C 10s, 69°C 20s, 72°C 1min
    Final extension 72°C 10min
    End at 4°C
