Protocols / A simple method for DNA isolation

A lot of people don’t know that our food contains genetic material. Therefor we performed a DNA isolation from bananas with housekeeping articles to educate interested passerby.

Material:

BananasTap water
Table saltDishwashing liquid as detergent
Washing agent with proteasesHot water (60 °C)
IceTeaspoon
TablespoonFork
Isopropyl (alcohol) (cooled)Coffee filter
Funnel50 mL vessels


Test execution:

  • preparation of extraction solution:
    1. 1 teaspoon table salt
    2. 1 tablespoon dishwashing liquid
    3. 10 drops washing agent
    4. Fill up to 100 mL with tap water
    5. Mixing by shaking
  • Homogenization of a banana with the fork
  • Transfer of 3 teaspoons homogenized banana into a 50mL reaction vessel with 30 mL extraction solution.
  • 15 min incubation at 60 °C
  • 5 min chilling on ice
  • Filter the mixture into a new 50 mL vessel.
  • Overlay the filtered solution with some iosporyl (alcohol)
  • Wait for 10 min
  • DNA is soaring into the phase of isopropyl (alcohol)
