The accuracy of each ncAA sythetase/tRNA pair was measured by comparing the GFP production of pStG/pFRY strains in (+/-) ncAA conditions. Strains containing pFRY and pStG plasmids were grown in the presence and absence of the corresponding non-cannonical amino acid.
