B0015, pSB1A3, pSB1C3, I746104, K081009, K0133222, S03156, E0040 and B0034 was inoculated from the glycerol stocks.
 
Digested with enzymes
 
S03156 was ligated to B0015, E0040 was ligated to B0015 and K081009 was ligated to B0015.
 
Transformed to E. coli DH5α
